This is the basic function PRS-IHC target array

Black / white targets to set the imaging white balance optimally to the slide
A row of rabbit protein gradient density 2D array from 10-100%
A row of mouse protein gradient density 2D array from 10-100%
Two targets composed of mouse and rabbit in a 3D matrix structure
The underlaying basis for using the secondary stain accumulative results as a density ruler.

Advanced PRS

In addition to the Essential targets there are:
Two mouse and rabbit targets to support a second round of primary antibody reagents.
Comparing the 50% 2D target of the gradient array with the second level 50% targets and applying
the difference to the created 3D array allows the antigen density scales to be created.
The antigen retrieval measure, ARM, targets are 2D and 3D deposits of a 50:50 mix of mouse and
rabbit. These show the action of the antigen retrieval as it assaults the targets seeking to
remove the formaldehyde fixation. The values of the two targets are
used to generate the ARM value used in QC of the antigen retrieval process.

The full function PRS-IHC includes up to ten primary antigen targets.
There are two different versions that can be supplied: 100-different antigens at 100% concentrations or 10-antigen pairs, where each pair is 80 and 40% concentrations.
The primary targets are used to modify the secondary 3D scale down should the primary not be capable of fully supporting the antigen demand. Such a situation is only expected during the development of a new antibody reagent.

Hematoxylin is not a stain by itself

Auto Pap Smear

Pap stains incorporate a second stain component that interacts with the not fully reacted hematoxylin stain: The Phosphotungstic acid binds to fibrin & collagen, but will also bind to unreacted mordanted hematein.

The result is that the hematoxylin stain now has a larger impact than in just H&E alone. Since there is not an IHC follow up, the risk of a false negative diagnosis is a serious issue.

To ensure consistent diagnostic determination the hematoxylin stain reagent must be held to a consistent concentration, pH, and reactivity.

Using digital imaging, the image can be normalized by using the PRS-PAP target information to compensate for hematoxylin drift

The PRS-PAP utilizes chemical targets that uniquely fully react with a single stain component so that each stain component can be objectively evaluated.

In a QC modality, the PRS-PAP validates the stain reagents are working normally.
